१९३० तक कुनैन मलेरिया की रोकथाम के लिए एकमात्र कारगर औषधि थी, बाद में एंटी मलेरिया टीके का प्रयोग भी इससे निपटने के लिए किया जाने लगा।

मलेरिया के विरूद्ध पहला प्रभावी उपचार सिनकोना वृक्ष की छाल से किया गया था जिसमें कुनैन पाई जाती है।

कुनैन पराबैंगनी प्रकाश संवेदी होती है, व सूर्य के प्रकाश से सीधे संपर्क में फ़्लुओरेज़ हो जाती है।

कुनैन का सर्वाधिक प्रचलित एवं मान्य हाइपोथीसिस इसके निकट संबंधी और पूर्ण अध्ययन किये गए क्विनोलिन ड्रग क्लोरोक्वीन पर आधारित है।

परन्तु छाल से कुनैन को १८२० तक अलग नहीं किया जा सका।

मूल शुद्ध रूप में कुनैन एक सफेद रंग का क्रिस्टल युक्त पाउडर होता है, जिसका स्वाद कड़वा होता है।

सिनसिनाटी मास्टर्स कुनैन (, ) एक प्राकृतिक श्वेत क्रिस्टलाइन एल्कलॉएड पदार्थ होता है, जिसमें ज्वर-रोधी, मलेरिया-रोधी, दर्दनाशक (एनल्जेसिक), सूजन रोधी गुण होते हैं।

इसके बाद कुनैन देने से मलेरिया और उपदंश दोनों काबू में आ जाते थे।
